In 1882 Franz Ziehl and Friedrich Neelsen demonstrated that certain bacteria stained with a hot phenol-fuchsin solution resisted decolorization with acid alcohol; these organisms were identified as acid-fast. Acid-fastness describes the physical property of certain bacteria's resistance to decolorization by dilute acids and/or alcohol based decolorization solutions. Acid-fast techniques are useful in identifying acid-fast organisms, mainly mycobacteria such as Mycobacterium tuberculosis, responsible for tuberculosis (TB), and Mycobacterium leprae responsible for leprosy. Acid-fast organisms are difficult to stain using standard stains, such as Gram's stain, due to the large amounts of lipids on the acid-fast organism's wall that resist these types of stain.

In 1915, JJ Kinyoun modified the Ziehl-Neelsen's technique so that the heating of the phenol-fuchsin solution was no longer required. During the past century, the Ziehl-Neelsen and Kinyoun methods have been modified by many practioners, but the basic process remains relevant today.

MasterTech's AFB Stain Kit
American MasterTech's AFB Stain Kitis suitable, in lieu of and comparable to the Kinyoun's AFB and Ziehl-Neelsen procedures for staining all mycobacteria, including Tubercle Bacillus. The MasterTech AFB Stain Kit differs from those procedures in that Advanced Carbol Fuchsin™ (ACF) is used for bacterial staining instead of a standard Carbol Fuchsin, and Light Green is used as a counterstain instead of Methylene Blue. In comparison to standard Carbol Fuchsins, MasterTech AFB Stain Kit requires no heat and much shorter staining times by combining three types of fuchsin dyes. Light Green is a much better counterstain than Methylene Blue, as it provides higher contrast against the red stained bacteria.

For labs with a fluorescence capable microscope, the AFB Modified Truant's Fluorescent Stain Kit provides a much greater contrast than standard light microscope procedures. The AFB Modified Truant's Fluorescent Stain Kit stains for mycobacteria (acid fast bacteria) in as little as 10 minutes. Auramine O-Rhodamine B serves as the primary stain, producing fluorescent yellow to orange bacteria against a black background produced by Eriochrome Black T.
