Alcian Blue stain is a catonic, water-soluble, phthalocyanin basic dye that's colored blue due to the presence of copper in its' molecule. When Alcian Blue Stain is used in an aqueous solution, it reacts to compounds with anionic groups, such as: mucin, mucosubstances, and DNA. It is possible that Alcian Blue forms salt linkages that act as the binding force, but this theory is not in complete agreement. American MasterTech offers four different types of Alcian Blue Stains: Alcian Blue Stain - pH 1.0, Alcian Blue Stain - pH 2.5, Alcian-PAS, and Alcian Blue Concentrate.

With its lower pH value, Alcian Blue Stain - pH 1.0 stains strongly sulfated mucins (mucosubstances). Alcian Blue - PAS
The Alcian Blue-PAS Stain Kit is used to differentiate neutral and acidic mucosubstances by staining the acidic mucosubstances blue using the alcian blue technique and the neutral mucosubstances pink with the PAS reaction. The MasterTech Alcian Blue-PAS Stain Kit does all this in just 60 minutes. The Alcian Blue-PAS Stain Kit contains a specially formulated Schiff's solution, eliminating the Sulfurous rinse step that is commonly found in similar stain kits on the market. This reduces stain procedure time and lessens exposure to sulfite fumes. Alcian Blue and Schiff's serve as the primary stains, producing blue acid mucins, purple combined acid/neutral mucins and glycogen with blue nuceli counterstained by Modified Mayer's Hematoxylin.
