Karnovsky’s Fixative was first developed by MJ Karnovsky in 1965. Karnovsky’s Fixative is an excellent, fast penetration fixative used in Electron Microscopy. Though, mainly used in Electron Microscopy, Karnovsky’s Fixative is also used in the fixation and storage of lung tissue used in animal inhalation.

Karnovsky’s Fixative was originally made from paraformaldehyde, 1M Sodium Hydroxide, 50% Glutaraldehyde, and 0.2M Cacodylate Buffer. pH 7.4. However, most Karnovsky’s Fixatives made today are of a modified version. The original Karnovsky’s Fixative original formulation was very hypertonic, forcing most technicians to dilute Karnovsky’s at half strength.
