The Oil Red O Method is a common staining technique used for demonstrating lipids in tissue. It is performed on fresh frozen sections, as alcohol and xylene dissolves the lipids that are in the tissue. Oil Red O has largely replaced many Sudan Dyes that were used for demonstrating lipids, such as Sudan III and Sudan IV, because Oil Red O provides a deeper red color that is easier to see.

The Oil Red O stain can be helpful in determining a number of pathologies such as detecting fat occurring in irregular places. When a bone fracture or a crushing injury occurs to places in the body containing fat, the fat can be released into circulation and cause a fatty embolism. Oil Red O stain can determine if the embolism is the cause of death. When structures that contain fat, such as cell membranes or myelin, go through a degenerative change, cells and fat may combine into fat droplets that are detectable by the Oil Red O stain. Oil Red O stain is also useful in detecting tumors that have developed from fat cells.
